چکیده

The development of a novel, sensitive enzyme-linked immunoassay (ELISA) for the detection of human lipocortin 1 [ I ] has enabled examination of the corticosteroid inducibility of this protein. Lipocortin 1 (molecular mass 37 kDa) has recently been the subject of intense investigation to substantiate early claims that it mediates the anti-inflammatory effects of corticosteroids by antagonising the activity o f phospholipase A,. Although it has clearly been shown to have phospholipase A, inhibitory activity in vitro [2, 31, it remains uncertain whether this is a direct enzyme inhibition or due to an interaction with substrate [4]. Recombinant lipocortin 1 has recently been shown to mimic the effect of corticosteroid on carrageenan-induced paw oedema in rats IS], but there is no direct evidence that the protein can be induced in vivo by corticosteroid in man, although the promoter region of the DNA sequence does possess potential corticosteroid receptor binding sites [6]. A bolus 100 mg intravenous dose of hydrocortisone was given to eight healthy male volunteers (median age 35 years) in an attempt to monitor changes in lipocortin 1 levels in peripheral blood leucocytes. Each experiment was begun at 13:OO h to correspond with the daily trough of endogenous circulating cortisol. Heparinized blood samples were taken before steroid administration and frequently over the course of the ensuing 240 min. As a control, an identical procedure was carried out on two subjects, the steroid infusion being replaced by placebo. Leucocytes were separated by flotation on a FicollHypaque M-85 [ 5 1% (w/v) meglumine diatrizoate, 26% (w/ v ) sodium diatrizoate gradient. Plasma was also collected and

Ganoderma Lucidum Induces the Expression of CD40/CD86 on Peripheral Blood Monocytes

Background: The major immuno-modulating effects of Ganoderma lucidum include mitogenicity and activation of immune effector cells such as T cells, macrophages and natural killer cells resulting in the production of cytokines. Objective: The purpose of this study was to evaluate the expression of CD40 and CD80 by G. lucidum-treated human peripheral blood mononuclear cells. Methods: Monocytes wer...

Arteether Exerts Antitumor Activity and Reduces CD4+CD25+FOXP3+ T-reg Cells in Vivo

Background: Chemo-immunotherapy is one of the new achievements for treatment of cancer, by which the success of anti-cancer therapy can be increased. In vitro studies have been shown that Arteether (ARE) induces apoptosis in tumor cells, but not in normal cells. Objective: To investigate the cytotoxic and immunomodulatory properties of Arteether in-vivo and in-vitro. Methods: In this study, we ...

Evaluation of proliferation and cytokines production by mitogen-stimulated bovine peripheral blood mononuclear cells

This in vitro study was conducted to evaluate lymphocyte blastogenic and cytokine production by bovine peripheral blood mononuclear cells (PBMCs) stimulated with phytohemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A (Con A) mitogens, by using tetrazolium salt and ELISA tests, respectively. The results presented that  Interleukin-2 (IL-2), IL-4, IL-5, IL-10, IL-17 and IFN-...

Changes in some pro-and anti-inflammatory cytokines produced by bovine peripheral blood mononuclear cells following foot and mouth disease vaccination

Interleukin (IL)-17 is exclusively produced by CD4 helper T-cells upon activation. It most often acts as a pro-inflammatory cytokine, which stimulates the release of pro-inflammatory cytokines IL-6, IL-8, TNF-α, and granulocyte-macrophage colony-stimulating factor (GM-CSF). In this study, we studied the in-vitro IL-17 response to specific antigens and a variety of mitogens and compared the IL-1...

Effect of 900 MHz Electromagnetic Radiation on the Induction of ROS in Human Peripheral Blood Mononuclear Cells

Background, Despite numerous studies over a decade, it still remains controversial about the biological effects of RF EMF emitted by mobile phone telephony. Objective, Here we investigated the effect of 900 MHz GSM on the induction of oxidative stress and the level of intracellular reactive oxygen species (ROS) in human mononuclear cells, monocytes and lymphocytes as defence system cells. Metho...
